Across literature and commercial listings, the substance appears under several names, including bremelanotide, PT-141, and various brand designations. A CAS registry number provides a unique identifier for the compound, aiding retrieval from chemical databases. Synonym lists may also contain early development codes and descriptive chemical names. When consulting sources, researchers need to confirm that the cited names refer to the same molecular structure to avoid confusion. Naming consistency matters especially when comparing analytical data across publications.
PT-141 is a synthetic cyclic heptapeptide whose development code is bremelanotide. It belongs to the class of melanocortin receptor agonists and acts by mimicking endogenous peptide hormones. The compound originated from research on melanotan II, where investigators exploring derivatives found distinct pharmacological features. Unlike the parent compound, PT-141 showed effects on pathways related to sexual desire and function in early studies, prompting its development as a separate candidate. Its molecular design aimed to separate receptor activity from pigmentary effects.
Outside the approved product, bremelanotide circulates as a research chemical sold by peptide vendors, often labelled PT-141. Such material is not manufactured under pharmaceutical quality standards, and independent testing has repeatedly found content that differs from the label. Analytical certificates supplied with a purchase are not strong evidence of purity because they are usually generated by the seller. Online discussion tends to blur the distinction between the approved drug and unregulated powder, which complicates interpretation of reported experiences.
PT-141 is the original development code for bremelanotide, a synthetic peptide first studied as a potential tanning and sexual-response agent in the 1990s. Researchers at a small American biotechnology firm designed it as a shortened analogue of melanotan II, which itself came from work on alpha-melanocyte-stimulating hormone. Early screening focused on pigmentation, but behavioural observations in animal models redirected attention toward sexual motivation. That shift made PT-141 one of the first melanocortin compounds investigated specifically for effects on desire rather than on skin colour.
Clinical development proceeded through two routes of administration. An intranasal formulation advanced first, but variable absorption and tolerability problems led to a switch to subcutaneous injection. The United States Food and Drug Administration approved the subcutaneous product in 2019 for hypoactive sexual desire disorder in premenopausal women. Marketing rights subsequently changed hands, and commercial availability has fluctuated since approval. Use in men, in postmenopausal women, and in combination with other agents remains outside the approved label.

Regulatory status varies by jurisdiction, where approved prescription products, compounded preparations and research-grade material are treated as distinct categories with different documentation requirements. Suppliers of research material commonly issue a certificate of analysis listing purity, identity and sometimes endotoxin content. Independent verification by a third-party laboratory is often recommended because self-reported figures are difficult to check. Literature discussions usually state the source, purity and storage conditions of the material used, since these details affect reproducibility. Analysts note that a reported purity figure does not by itself describe biological activity.
Lyophilised peptide is generally held below minus twenty degrees Celsius, protected from light and moisture, because hydrolysis and oxidation accumulate faster at ambient temperature. Once reconstituted, solutions are typically kept between two and eight degrees Celsius and used within a short window defined by the supplier. Repeated freeze-thaw cycles are avoided since they promote aggregation and loss of soluble material. Container material matters as well, because peptides adsorb to certain plastics and glass surfaces at low concentration. Stability figures supplied by a vendor apply only to the specific lot and buffer that were tested.
Identity and purity are usually established with reversed-phase high-performance liquid chromatography combined with mass spectrometry. A gradient of water and acetonitrile containing trifluoroacetic acid is a common mobile phase, and ultraviolet detection near 214 nanometres responds to the peptide backbone. Mass spectrometry confirms the expected molecular mass and helps reveal truncation or oxidation products. Purity is reported as a peak-area percentage, a figure that depends on the wavelength and gradient used, so values from different laboratories are not always directly comparable. Peptide mapping and amino acid analysis provide additional confirmation when required.
=== Cancer === Inflammation orchestrates the microenvironment around tumours, contributing to proliferation, survival and migration. Cancer cells use selectins, chemokines and their receptors for invasion, migration and metastasis. On the other hand, many cells of the immune system contribute to cancer immunology, suppressing cancer. Molecular intersection between receptors of steroid hormones, which have important effects on cellular development, and transcription factors that play key roles in inflammation, such as NF-κB, may mediate some of the most critical effects of inflammatory stimuli on cancer cells. This capacity of a mediator of inflammation to influence the effects of steroid hormones in cells is very likely to affect carcinogenesis. On the other hand, due to the modular nature of many steroid hormone receptors, this interaction may offer ways to interfere with cancer progression, through targeting of a specific protein domain in a specific cell type. Such an approach may limit side effects that are unrelated to the tumor of interest, and may help preserve vital homeostatic functions and developmental processes in the organism. There is some evidence from 2009 to suggest that cancer-related inflammation (CRI) may lead to accumulation of random genetic alterations in cancer cells.
=== Initial adoption === Due to the influence tire pressure has on vehicle safety and efficiency, tire-pressure monitoring (TPM) was adopted by the European market as an optional feature for luxury passenger vehicles in the 1980s. The first passenger vehicle to adopt TPM was the Porsche 959 in 1986, using a hollow spoke wheel system developed by PSK. In 1996 Renault used the Michelin PAX system for the Scenic and in 1999 PSA Peugeot Citroën decided to adopt TPM as a standard feature on the Peugeot 607. The following year (2000), Renault launched the Laguna II, the first high volume mid-size passenger vehicle in the world to be equipped with TPM as a standard feature. In the United States, TPM was introduced by General Motors for the 1991 model year for the Corvette in conjunction with Goodyear run-flat tires. The system uses sensors in the wheels and a driver display which can show tire pressure at any wheel, plus warnings for both high and low pressure. It has been standard on Corvettes ever since.
John Hurrell Crook studied the behaviour of weaverbirds and demonstrated the links between ecological conditions, behaviour, and social systems. Principles from economics were introduced to the study of biology by Jerram L. Brown in his work on explaining territorial behaviour. This led to more studies of behaviour that made use of cost-benefit analyses. The rising interest in sociobiology also led to a spurt of bird studies in this area. The study of imprinting behaviour in ducks and geese by Konrad Lorenz and the studies of instinct in herring gulls by Nicolaas Tinbergen led to the establishment of the field of ethology. The study of learning became an area of interest and the study of bird songs has been a model for studies in neuroethology. The study of hormones and physiology in the control of behaviour has also been aided by bird models. These have helped in finding the proximate causes of circadian and seasonal cycles. Studies on migration have attempted to answer questions on the evolution of migration, orientation, and navigation. The growth of genetics and the rise of molecular biology led to the application of the gene-centered view of evolution to explain avian phenomena. Studies on kinship and altruism, such as helpers, became of particular interest. The idea of inclusive fitness was used to interpret observations on behaviour and life history, and birds were widely used models for testing hypotheses based on theories postulated by W. D. Hamilton and others.
He also co-founded Animal Bioscience in 2017 along with his brother Nick. The company focuses on small molecule-based therapy for the pet industry. In the same year he founded Life Biosciences to develop gene therapies aimed at age-related disease. In 2022, Metro Biotech successfully urged the FDA to take actions to take NMN off the market as a supplement because Metro Biotech had registered NMN in investigational new drug applications. The following year, he co-founded Tally Health, a supplement company with a stated goal to "change the way we age" at the cellular level.
transferase Any of a class of enzymes which catalyze the chemical transfer of a functional group or substituent from one molecule to another. For example, acetyltransferases catalyze the movement of an acetyl group in a process known as acetylation; methyltransferases catalyze the movement of one or more methyl groups in a process known as methylation.

== Diagnosis == LS should be suspected in children or adults with distinctive physical features listed above, extremely elevated serum hGH concentrations despite low serum IGF-1 levels. A failure of IGF-1 to increase in response to exogenous hGH (IGF-1 stimulation test) is diagnostic for LS. The gold standard for confirming a diagnosis of LS is to perform a genetic analysis with PCR to identify the precise molecular defect in the GH receptor gene. Other laboratory abnormalities include GHBP (growth hormone binding protein) levels being low in cases with mutations in the extracellular domain of the GH receptor and normal in cases with mutations in the intracellular domain. Low serum levels of IGFBP are non-diagnostic for LS.
== Mechanisms == Huntingtin protein (HTT) interacts with over 100 other proteins, and appears to have multiple functions. The behavior of the mutated protein (mHTT) is not completely understood, but it is toxic to certain cell types, particularly brain cells. Early damage is most evident in the subcortical basal ganglia, initially in the striatum, but as the disease progresses, other areas of the brain are also affected, including regions of the cerebral cortex. Early symptoms are attributable to functions of the striatum and its cortical connections—namely control over movement, mood, and higher cognitive function. DNA methylation also appears to be changed in HD. In 2025, scientists affiliated with Harvard and MIT published a study examining the mechanism behind the onset of symptoms. They found that the length of the repeated trinucleotide sequence increases with age due to accumulated errors in DNA mismatch repair processes after transcription, and that this becomes toxic once the sequence expands beyond 150 repeats.
In the early part of 1982, Carrey reportedly performed for The Tonight Show bookers Jim McCawley and Bud Robinson as part of the program's audition process for stand-up comic spots. However, rather than being booked on the show, Carrey got advised to further hone his act, so he went back home to the Toronto area where he had already built a significant following. Touring venues throughout North America as the opening act for Rodney Dangerfield, Carrey made a stop at home in Toronto on June 19, 1982, performing two sold-out shows at Massey Hall.
== Adverse effects == The most common reported adverse effects were nausea (19%), headache (20%), and implant site reactions (21%). Uncommon but potentially serious hypersensitivity reactions including anaphylaxis were also reported. Afamelanotide was also found to sometimes induce darkening of certain pre-existing skin conditions, so regular full body skin examinations every 6 months is often recommended. Other common adverse effects reported during the clinical trials include back pain, upper respiratory tract infections, melanocyte naevus, decreased appetite, migraine, dizziness, weakness, fatigue, lethargy, sleepiness, hot flashes, abdominal pain, diarrhea, vomiting, flushing, development of warts, spots, and freckles, and itchy skin (between 1% and 10% of people). Uncommon and rare adverse effects include cystitis, folliculitis, gastrointestinal infections, hypersensitivity reactions, changes in appetite, depression, insomnia, balance disorders, lethargy, restless leg syndrome, syncope, photophobia, presbyopia, tinnitus, confusion, palpitations, hypertension, hypercholesterolaemia, and weight gain.
lipid Any of a heterogeneous class of organic compounds, including glycerides (fats), waxes, sterols, and some vitamins, united only by their amphipathic or hydrophobic nature and consequently their very low solubility in water. Some lipids such as phospholipids tend to form lamellar structures or micelles in aqueous environments, where they serve as the primary constituents of biological membranes. Others such as fatty acids can be metabolized for energy, have important functions in energy storage, or serve as signaling molecules. Colloquially, the term "lipids" is sometimes used as a synonym for fats, though fats are more correctly considered a subclass of lipids.

, regardless of secondary structure. The modern versions of these libraries as used in most software are presented as multidimensional distributions of probability or frequency, where the peaks correspond to the dihedral-angle conformations considered as individual rotamers in the lists. Some versions are based on very carefully curated data and are used primarily for structure validation, while others emphasize relative frequencies in much larger data sets and are the form used primarily for structure prediction, such as the Dunbrack rotamer libraries. Side-chain packing methods are most useful for analyzing the protein's hydrophobic core, where side chains are more closely packed; they have more difficulty addressing the looser constraints and higher flexibility of surface residues, which often occupy multiple rotamer conformations rather than just one.
The antibody selectively binds to protein A/G, so a high level of purity (generally >80%) is obtained. The generally harsh conditions of this method may damage easily damaged antibodies. A low pH can break the bonds to remove the antibody from the column. In addition to possibly affecting the product, low pH can cause protein A/G itself to leak off the column and appear in the eluted sample. Gentle elution buffer systems that employ high salt concentrations are available to avoid exposing sensitive antibodies to low pH. Cost is also an important consideration with this method because immobilized protein A/G is a more expensive resin. To achieve maximum purity in a single step, affinity purification can be performed, using the antigen to provide specificity for the antibody. In this method, the antigen used to generate the antibody is covalently attached to an agarose support. If the antigen is a peptide, it is commonly synthesized with a terminal cysteine, which allows selective attachment to a carrier protein, such as KLH during development and to support purification. The antibody-containing medium is then incubated with the immobilized antigen, either in batch or as the antibody is passed through a column, where it selectively binds and can be retained while impurities are washed away. An elution with a low pH buffer or a more gentle, high salt elution buffer is then used to recover purified antibody from the support.
=== Hepatic Function === The liver stores excess glucose in the form of glycogen after a meal, in response to insulin. Between meals (or during fasting), the liver is stimulated to break down this glycogen to release glucose (glycogenolysis) and also synthesizes new glucose (gluconeogenesis); this glucose is released into the bloodstream to maintain normal function of the brain and other organs that burn glucose for energy. Glycogenolysis and gluconeogenesis are stimulated by hormones such as glucagon that activate the cyclic AMP pathway in liver hepatocytes, and this cAMP promotes activation of metabolic enzymes leading to glucose production and release; asprosin appears to utilize this same system of control. Asprosin was reported to stimulate glucose release from hepatocytes, and plasma levels of asprosin in obese high-fat-fed mice have been reported to nearly double. However, in a study in 2019, a pharma replication group reported their inability to replicate these two key observations using recombinant asprosin, suggesting that issues with reagent purity may have been responsible for the effect observed in the initial asprosin study. Nevertheless, a third group reported in 2019 that they had identified the liver receptor for asprosin, OR4M1, an olfactory receptor family GPCR, and showed that plasma asprosin levels increased with fasting and with diet-induced obesity, and confirmed asprosin's effect on stimulation of hepatic glucose production, replicating all facets of the original study.
=== Canada === The Mexican Army curtailed the ability of the Mexican drug cartels to move cocaine inside the U.S. and Canada, prompting an upsurge in gang violence in Vancouver in 2009, where the cocaine price has increased from $23,300 to almost $39,000 per kilo as the Canadian drug markets experienced prolonged shortages. As evidence of this pressure, the U.S. government stated the amount of cocaine seized on U.S. soil dropped by 41 percent between early 2007 and mid-2008. Since 2009, Vancouver has become the Mexican cartels' main center of operations in Canada.
Mass spectrometry is used to separate and measure distinct isotopes present in a sample. Archaeologists typically employ isotope ratio mass spectrometers or IRMSs, consisting of an inlet system, ion source, mass analyser and multiple ion detectors. The sample is usually introduced into the mass spectrometer as a gas, with oxygen and carbon being introduced as carbon dioxide. Strontium is too unstable to be easily handled in gas form, instead, it is evaporated and ionised in a vacuum. This use of a solid source is referred to as thermal ionisation mass spectrometry or TIMS. More recently, strontium isotopes have been at the centre of discussion and investigation into the use of laser ablation inductively coupled mass spectrometry (ICP-MS), which is also of interest due to its less invasive nature. Electron bombardment ionises the gas, allowing the molecules to be focused into a beam which is then split by mass into smaller beams - forming a "mass spectrum". The relative intensities of the different beams is then measured in the ion collector and relayed as isotope ratios.
