Bremelanotide is a moderately large peptide with a molecular mass near 1025 daltons. In lyophilised form it appears as a white to off-white powder and is freely soluble in water and other polar solvents. The intact lactam ring is essential for receptor affinity, while linearised fragments bind far more weakly. Solutions are sensitive to extremes of pH and to prolonged exposure to light and heat, so handling typically involves buffered conditions and cold storage. Its short plasma half-life reflects rapid distribution and clearance rather than chemical breakdown inside the vial.
Development of the peptide passed through several delivery formats, including an intranasal version tested in early trials and an injectable version that entered later clinical study. Regulatory approval for a subcutaneous product in the United States was granted in 2019 after review of controlled trials in premenopausal women. Outside the clinic, the compound circulates in research and non-pharmaceutical markets under its code name, where identity and purity vary considerably between suppliers. Synonyms appearing across technical literature include bremelanotide, PT-141, and Palatin 141.
Published discussion sits at the intersection of peptide chemistry, neuroendocrinology and sexual medicine. Trial reports emphasise change scores on validated instruments, while mechanistic papers focus on hypothalamic circuits and receptor selectivity. Because placebo response in this field is large, effect sizes are usually reported with confidence intervals rather than as isolated averages. Reviews note that female and male data sets are not interchangeable and should be read separately. Diagnostic terminology has been revised over time, which complicates comparison between older and newer studies.
Bremelanotide is a synthetic cyclic heptapeptide that acts on a family of G-protein-coupled receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide associated with pigmentation and several central signalling pathways. A lactam bridge constrains the ring and slows enzymatic breakdown, which distinguishes it from the linear parent molecule. Research interest moved over time from pigment biology toward central nervous system effects, particularly circuits connected to sexual desire. Parenteral delivery is used because oral bioavailability is poor.
Early clinical work used an intranasal formulation, and later programmes switched to subcutaneous delivery for more consistent absorption. A subcutaneous product received regulatory approval in the United States in 2019 for premenopausal women with acquired, generalised hypoactive sexual desire disorder. Approval followed phase 3 trials in which active treatment separated from placebo on desire and distress measures, though the average difference was modest. Labeling carries a caution about transient blood pressure elevation, so cardiovascular history is assessed before prescribing. Questions about durability of benefit beyond several months remain open.

Certificate of analysis documents usually report purity percentage, molecular weight, and appearance. Researchers verify identity by comparing observed and theoretical mass values. Chain-of-custody records and batch numbers help trace material from source to experiment. Independent verification of supplier claims is considered good practice when material identity is critical to a study. Records should include the storage history of each aliquot, and unexpected deviations in appearance or solubility warrant re-testing before use.
Lyophilized peptide material is generally stored at -20 °C or below to limit degradation, while reconstituted solutions are less stable and are typically kept refrigerated and protected from light. Repeated freeze-thaw cycles can accelerate aggregation and should be minimized. Stability for any specific lot depends on purity, moisture content, and packaging. Handling in a temperature-controlled environment reduces variability across replicates, and exposure to ambient humidity during weighing can introduce error. Aliquotting reduces the number of times a stock container is opened.
After subcutaneous administration, plasma concentrations rise within roughly thirty minutes and the elimination half-life is short, on the order of two to three hours. Reported physiological responses include transient increases in blood pressure and nausea, which tended to diminish with repeated dosing in trial settings. Because the peptide clears quickly, effects are not expected to persist long after a dose. Absorption from non-injected routes is poorly characterised, and nasal delivery produced variable plasma levels in older work.
Clinical research typically uses randomised, double-blind, placebo-controlled designs. The most common primary endpoint is the desire domain score of the Female Sexual Function Index, sometimes paired with a distress measure. Secondary outcomes include arousal, satisfaction, and event-based counts of satisfying sexual episodes. Across trials, average improvements are modest and individual responses vary widely. Whether benefits persist beyond a few months, and whether they depend on baseline hormone status, remain open questions rather than settled findings.
Melanocortin receptors form a family of five G-protein-coupled receptors designated MC1 through MC5. Bremelanotide binds most strongly at MC4R and MC1R, with weaker activity reported at MC3R and MC5R. MC4R is expressed in hypothalamic nuclei that coordinate energy balance and aspects of sexual behaviour. The prevailing interpretation is that central MC4R activation, rather than peripheral vascular effects alone, drives the reported changes in desire. This account remains partly inferential, since direct receptor-level measurement in living humans is not practical.
=== Medical === Tracking expression has allowed for multiple investigations into the progression of diseased cells. Reporter genes have shown to provide critical insight into genes upregulated in cancer regulatory pathways as well as the identification into oncogenes and tumor suppressor genes. These have been used for further research into the development of therapeutics to stop further disease progression and metastasis. Gene therapy has also been tracked through the use of reporter genes. This allows for the monitoring of gene therapy vectors to see if they are achieving intended results as well as to monitor patient safety for short and long term periods. Therapeutics developed have benefited from the use of reporter genres such as a dual-reporter system developed for CRISPR/Cas9 models to monitor progression and success and benefits of being gene editing tools.
=== Ion pair extraction === It is possible by careful choice of counterion to extract a metal. For instance, if the nitrate concentration is high, it is possible to extract americium as an anionic nitrate complex if the mixture contains a lipophilic quaternary ammonium salt. An example that is more likely to be encountered by the 'average' chemist is the use of a phase transfer catalyst. This is a charged species that transfers another ion to the organic phase. The ion reacts and then forms another ion, which is then transferred back to the aqueous phase. For instance, the 31.1 kJ mol−1 is required to transfer an acetate anion into nitrobenzene, while the energy required to transfer a chloride anion from an aqueous phase to nitrobenzene is 43.8 kJ mol−1. Hence, if the aqueous phase in a reaction is a solution of sodium acetate while the organic phase is a nitrobenzene solution of benzyl chloride, then, when a phase transfer catalyst, the acetate anions can be transferred from the aqueous layer where they react with the benzyl chloride to form benzyl acetate and a chloride anion. The chloride anion is then transferred to the aqueous phase. The transfer energies of the anions contribute to that given out by the reaction. A 43.8 to 31.1 kJ mol−1 = 12.7 kJ mol−1 of additional energy is given out by the reaction when compared with energy if the reaction had been done in nitrobenzene using one equivalent weight of a tetraalkylammonium acetate.
On 22 February 2026, El Mencho was killed by the Mexican military in Tapalpa, Jalisco. Widespread violence erupted in Mexico as a result of retaliation attacks from the CJNG. Mexico City-based security consultant David Saucedo informed CNN that due to El Mencho's son Rubén Oseguera González ("El Menchito") being jailed in the United States, the line of succession "has been broken," with no clear family successor. The arrest of El Menchito would leave only one brother and El Mencho's stepson Juan Carlos Valencia Gonzalez ("El Pelón"), though El Pelon lacked "influence among other cartel commanders." Saucedo stated that was "likely that one of four or five top commanders will emerge as the next leader," and named CJNG leaders Ricardo Ruiz Velasco ("El Doble R"), Audias Flores ("El Jardinero" or "The Gardener"), Hugo Mendoza Gaytan ("El Sapo") and a fourth figure – El Mencho's former head of security – about whom little is known, as the most likely candidates to succeed El Mencho. In addition it was soon afterwards revealed by El Pais that El Mencho's brother Abraham, who was arrested in Mexico in February 2025, was actually in fact still "in the hands of Mexican authorities." The U.S. Director of National Intelligence's (DNI) National Counterterrorism Center has stated that El Pelón was the "de facto second-in-command" to El Mencho, with $5 million bounties being issued each for El Pelon and El Jardinero.
=== Kinetic isotope effects === Kinetic isotope effect (KIE) occurs in irreversible reactions, such as methanogenesis, and can deviate methane clumped isotopologue composition from its thermodynamic equilibrium. Normally, KIE significantly drives
AM-1235 metabolism differs only slightly from that of JWH-018. AM-1235 N-dealkylation produces 1-fluoropentane instead of pentane or other simple alkanes in general. It had been speculated that the fluoropentane might function as an alkylating agent or be further metabolized into toxic fluoroacetic acid. This is not true, however, as fluoroalkanes do not act as alkylating agents under normal conditions and odd-length fluoroalkane chains metabolize into substantially less toxic fluoropropanoic acid.

=== March === 8 March The report of a 20-year survey by The Botanical Society of Britain and Ireland revealed that 56% of Ireland's native plant species are in decline due to habitat loss, altered grazing pressure, and degradation (re-seeding, over-fertilising, nitrogen deposition, herbicides, soil drainage, mineral enrichment), rather than rising temperatures whose botanical effects – so far – are minor. By contrast, 80% of non-native plants introduced into Ireland since 1500 have increased. The Taoiseach Leo Varadkar marked International Women's Day by announcing the Government's intention to hold a referendum to enshrine gender equality in the Constitution by amending Articles 40 and 41. 13 March – President of the United States Joe Biden announced that he will visit Ireland, north and south, in April for the 25th anniversary of the Good Friday Agreement (signed on 10 April 1998). It was reported two weeks later that Biden will arrive in Belfast on 11 April, then he will be based in Dublin from 12 to 15 April from where he will make trips to other parts of the country. Former US president Bill Clinton and his wife Hillary will visit Belfast the following week. 15 March – 46-year-old Stephen Silver was found guilty of the capital murder of Detective Garda Colm Horkan who was shot dead in Castlerea, County Roscommon in June 2020. 16 March – Ryan Tubridy announced that he would be stepping down as the presenter of The Late Late Show after 14 years, with his last show in May.
== In literature == Michael Morpurgo's children's book The Sleeping Sword (2002, ISBN 978-0749748524) was inspired by the discovery of the sword on Bryher, combining it with Arthurian legend. He dedicated the book to the people of Bryher, where he has spent many holidays.
The Buran could be fitted with four Saturn AL-31 turbofan engines and a fuel tank in its payload bay, allowing it to make its own atmospheric test flights, which began in November 1985. Also unlike the US Shuttle, it could be flown pilotlessly and landed automatically. Energia-Buran made only one orbital test flight in November 1988, but US counterintelligence baited the Soviets with disinformation about the heat shield design, and it was not reusable for repeated flight. Buran was the largest and most expensive Soviet program in the history of the Space Race, and was effectively canceled by the collapse of the Soviet Union in 1991, due to lack of funding. The Energia was also canceled at the same time, after only two flights.
Islam dominates in Pakistan, with about 96.35% of the population being Muslim. Pakistan ranks second globally in Muslim population, and is home to 10.5% of the world's Muslims. Karachi is the largest Muslim city in the world. The majority follow Sunni Islam, with a significant presence of Sufism, while Shia Muslims constitute a minority. Shias represent between 5–25%. The Shia population in Pakistan was estimated at 42 million in 2019. As of 2012, 12% of Pakistani Muslims self-identify as non-denominational Muslims. The Ahmadis are a minority, officially considered non-Muslims. Ahmadis face persecution, banned from calling themselves Muslims since 1974.
== Mechanism of action == Lebrikizumab blocks interleukin 13 (IL-13), a cytokine (cell-signalling protein) that is produced by a type of white blood cell called Th2 cells. IL-13 is thought to induce the expression of another signalling protein, periostin, by epithelial cells of the bronchi. Periostin in turn seems to partake in a number of asthma related problems, such as bronchial hyperresponsiveness, inflammation, and activation and proliferation of airway fibroblasts, which are involved in airway remodelling. This theory is supported by the fact that people with high periostin levels responded significantly better to lebrikizumab in the phase II study: the forced expiratory volume in 1 second (FEV1) was 8.2% higher than under placebo in this group (measured from the respective baselines), while low-periostin participants had 1.6% higher FEV1, and the average value for all participants was 5.5%. The FEV1 increase in low-periostin participants was not statistically significant.

Some of the steel transmitter towers remain, although the wooden receiver towers have all been demolished. The remaining towers have various new uses and in some cases are now protected as listed buildings by order of English Heritage. One such 360-foot-high (110 m) transmitter tower can now be found at the BAE Systems facility at Great Baddow in Essex, on the former Marconi Research Centre site. It originally stood at RAF Canewdon in Essex and was moved to Great Baddow in 1956. This is the only surviving Chain Home tower still in its original, unmodified form with cantilever platforms at 50 ft, 200 ft and 360 ft, and in 2019 was given Grade II listed status. Swingate transmitting station in Kent (originally AMES 04 Dover) has two original towers (three until 2010) which are used for microwave relay; the towers lost their platforms in the 1970s. RAF Stenigot in Lincolnshire has another, almost complete tower, without its top platforms; it is used for training aerial erectors. The only original Chain Home site which is still used as a military radar station is RRH Staxton Wold in North Yorkshire, although there are no remnants of the 1937 equipment as it was completely cleared and remodelled for the ROTOR replacement, the Linesman/Mediator system, in 1964. The 240-foot timber receiver towers were some of the tallest wooden structures ever built in Britain. Two of these wooden towers were still standing in 1955, at Hayscastle Cross. Unlike the transmitter tower pictured here, those at Hayscastle Cross were guyed.

=== Defence arguments === Letby's defence argued that she was a dedicated nurse working within a system that had failed, and that the prosecution's case relied on assumptions of deliberate harm combined with her presence during certain incidents. They contended that shortcomings in staffing and care on the neonatal unit were extensive and could not reasonably be attributed to a single individual. The defence also suggested that "extraordinary bleeding" in one infant might have been caused by medical equipment rather than deliberate injury. Counsel for the defence maintained that the prosecution had not presented sufficient evidence to support its theories of how the infants were harmed. They argued that the evidence for air embolism was too weak to sustain the allegation and noted that the term did not appear in Letby's internet search history. No independent medical experts were called by the defence. The only defence witness other than Letby was a plumber who testified that the unit had recurring plumbing problems, including sewage backing up into sinks, which he said required frequent call‑outs. The defence suggested that such hygiene issues could have contributed to the unit's elevated mortality rate. In later reporting, Dewi Evans acknowledged that pseudomonas had been detected in the unit's water supply, leading to several cases of pneumonia.
=== Etymology === Lactobacillic acid and Lactobacillussäure are translations of the English term 'lactobacillic acid', which scientists proposed when this fatty acid was discovered (1950) in a Lactobacillus species. In the German-language literature, the term lactobacillic acid is used more frequently than lactobacillus acid. As early as 1938, another group of researchers discovered an unusual fatty acid from the bacterium Agrobacterium tumefaciens (at that time called Bacterium tumefaciens or Phytomonas tumefaciens) was isolated and after the generic name as phytomonic acid. According to the knowledge of scientists at the time, this saturated fatty acid had the molecular formula C20H40O2. The proposed structure was a branched-chain fatty acid with a methyl group as a branch, methylnonadecanoic acid. However, K. Homann et al. were able to show in 1955 that this compound isolated from P. tumefaciens was in fact lactobacillic acid. According to them, the substance originally investigated had been contaminated.

PT-141 is a laboratory code for bremelanotide, a synthetic peptide that activates melanocortin receptors. It has been developed both as an injectable pharmaceutical product and as a research chemical. The two contexts differ sharply in how identity, purity, and dosing are controlled.
It is neither a steroid nor a naturally occurring human hormone. Bremelanotide is a manufactured peptide that mimics part of the melanocortin signalling system by binding receptor proteins. Its effects come from receptor activation, not from replacing a circulating hormone.
The internal lactam bridge locks the peptide into a defined shape, which raises receptor affinity and slows enzymatic breakdown. Linear versions of the same sequence are less stable and bind more weakly. Ring geometry is therefore central to how the molecule behaves in the body.
It is a synthetic cyclic peptide that activates melanocortin receptors. It is given by injection and was approved in the United States in 2019 for a specific low-desire diagnosis in premenopausal women. It is not a hormonal therapy.
