PT-141 is the research designation for bremelanotide, a synthetic cyclic heptapeptide developed as a melanocortin receptor agonist. Cyclisation of this kind improves resistance to enzymatic degradation relative to linear analogues. The compound was first explored for effects on melanin production, because several melanocortin receptors influence pigmentation. ==== Distribution ==== Atomoxetine is well-distributed and its volume of distribution is 0.85 to 1.02 L/kg in CYP2D6 extensive metabolizers and 2.25 L/kg in CYP2D6 poor metabolizers, with its distribution being equivalent to total body water. Atomoxetine and its metabolites show only limited partitioning into red blood cells. Atomoxetine crosses the blood–brain barrier, with this appearing to be due primarily to passive diffusion rather than active transport. The drug is not a substrate of P-glycoprotein. Animal studies have found that atomoxetine and/or its metabolites can cross the placenta, but fetal exposure was substantially lower than maternal exposure. Similarly, only a small amount of atomoxetine and/or metabolites were excreted in milk in animals. Atomoxetine shows high plasma protein binding of 98.7% and is primarily bound to albumin and to a lesser extent to α1-acid glycoprotein and immunoglobulin G (IgG). Atomoxetine's limitedly active metabolite N-desmethylatomoxetine is 99.1% bound to plasma proteins, whereas its active metabolite 4-hydroxyatomoxetine shows plasma protein binding of 66.6%, which is substantially less than that of atomoxetine itself.

Following the acquisition of Allergan plc generic division some assets have been sold off to meet US antitrust requirements. In June 2016 Indian pharmaceutical company Dr. Reddy's Laboratories Ltd bought 8 (ANDA) Abbreviated New Drug Applications for $350 million in cash. Also in June 2016 Teva sold two ANDAs to Indian pharmaceutical company Zydus Cadilla, strengthening its US portfolio. On June 21, 2016, American pharmaceutical company Amneal Pharmaceutical previously known as Impax Laboratories bought a portfolio of generic drugs from Teva Pharmaceutical Industries for about $586 million. On July 29, Cipla an Indian pharmaceutical company bought three products from Teva. Indian pharmaceutical company, Aurobindo, was in the race to buy some Teva assets. In October 2016, Teva has sold off part of its UK and Ireland generic business to Indian pharmaceutical company Intas Pharma for 600 million pounds (5083 crore Indian rupees) in cash. In August 2016, Australia pharmaceutical company Mayne Pharmaceutical bought a portfolio of drugs from pharmaceutical giant Teva Pharmaceuticals last year for $845 million Australian dollars.

== Medical use == Mebeverine is used to alleviate some of the symptoms of irritable bowel syndrome (IBS) and related conditions; specifically stomach pain and cramps, persistent diarrhoea, and flatulence. Historically data from controlled clinical trials have not found a difference from placebo or statistically significant results in the global improvement of IBS. However, a 2022 systematic review found that Mebeverine is an effective treatment option in IBS, with a good safety profile and low frequency of adverse effects. It has not been tested in pregnant women nor in pregnant animals so pregnant women should not take it; it is expressed at low levels in breast milk, while no adverse effects have been reported in infants, breastfeeding women should not take this drug.
== Contraindications == Due to its effects on blood pressure (generally a transient increase in systolic blood pressure by 6 mmHg, and diastolic blood pressure by 3 mmHg), bremelanotide is considered contraindicated in people with uncontrolled high blood pressure or cardiovascular disease. As long as bremelanotide is not used more than once in one day, it is not expected to cause more severe increases in blood pressure.

=== Mechanism of action === Transamination catalyzed by aminotransferase occurs in two stages. In the first step, the α amino group of an amino acid is transferred to the enzyme, producing the corresponding α-keto acid and the aminated enzyme. During the second stage, the amino group is transferred to the keto acid acceptor, forming the amino acid product while regenerating the enzyme. The chirality of an amino acid is determined during transamination. For the reaction to complete, aminotransferases require participation of aldehyde containing coenzyme, pyridoxal-5'-phosphate (PLP), a derivative of Pyridoxine (Vitamin B6). The amino group is accommodated by conversion of this coenzyme to pyridoxamine-5'-phosphate (PMP). PLP is covalently attached to the enzyme via a Schiff Base linkage formed by the condensation of its aldehyde group with the ε-amino group of an enzymatic Lys residue. The Schiff base, which is conjugated to the enzyme's pyridinium ring, is the focus of the coenzyme activity.
