Development of the peptide passed through several delivery formats, including an intranasal version tested in early trials and an injectable version that entered later clinical study. Regulatory approval for a subcutaneous product in the United States was granted in 2019 after review of controlled trials in premenopausal women. Outside the clinic, the compound circulates in research and non-pharmaceutical markets under its code name, where identity and purity vary considerably between suppliers. Synonyms appearing across technical literature include bremelanotide, PT-141, and Palatin 141.
PT-141 is the research designation for bremelanotide, a synthetic cyclic heptapeptide developed as a melanocortin receptor agonist. The peptide contains seven amino acids with an internal lactam bridge that constrains the backbone into a stable ring. Cyclisation of this kind improves resistance to enzymatic degradation relative to linear analogues. The compound was first explored for effects on melanin production, because several melanocortin receptors influence pigmentation. Its later association with sexual desire emerged from observations made during that early work, and the molecule became the subject of a separate development programme.
Clinical research typically uses randomised, double-blind, placebo-controlled designs. The most common primary endpoint is the desire domain score of the Female Sexual Function Index, sometimes paired with a distress measure. Secondary outcomes include arousal, satisfaction, and event-based counts of satisfying sexual episodes. Across trials, average improvements are modest and individual responses vary widely. Whether benefits persist beyond a few months, and whether they depend on baseline hormone status, remain open questions rather than settled findings.
Melanocortin receptors form a family of five G-protein-coupled receptors designated MC1 through MC5. Bremelanotide binds most strongly at MC4R and MC1R, with weaker activity reported at MC3R and MC5R. MC4R is expressed in hypothalamic nuclei that coordinate energy balance and aspects of sexual behaviour. The prevailing interpretation is that central MC4R activation, rather than peripheral vascular effects alone, drives the reported changes in desire. This account remains partly inferential, since direct receptor-level measurement in living humans is not practical.

Development began with intranasal formulations investigated for erectile dysfunction, but blood pressure elevation limited that route and prompted a switch to subcutaneous delivery. Clinical testing then shifted toward hypoactive sexual desire disorder in premenopausal women, and a subcutaneous product received United States approval in 2019. Later trials examined other populations with mixed results, and questions about effect size, durability and patient selection remain open in the peer-reviewed literature. Research interest continues in parallel with the broader melanocortin field, where several synthetic analogues are studied together.
PT-141 is the research code for bremelanotide, a cyclic heptapeptide derived from alpha-melanocyte-stimulating hormone. The molecule belongs to the melanocortin receptor agonist family and shows highest affinity for the MC4 receptor subtype, with weaker activity at MC1, MC3 and MC5. Its structure retains the core His-Phe-Arg-Trp sequence that defines melanocortin recognition, while cyclization and terminal modifications improve metabolic stability relative to the parent hormone. Early work classified the compound as a centrally acting agent rather than a peripherally acting vasodilator, which shaped subsequent development priorities.
Receptor activation in hypothalamic and limbic circuits is the mechanism most often cited for the observed effects on sexual desire. Signalling through MC4R couples to Gs proteins and raises intracellular cyclic AMP, which in turn modulates dopaminergic tone in reward-related pathways. Because the peptide reaches the central nervous system after subcutaneous administration, peripheral vascular changes are regarded as secondary rather than primary. The precise neural circuits that translate receptor occupancy into behavioural change remain incompletely mapped, and published accounts describe the pathway in general terms rather than as a fully resolved sequence.
The lactam ring that closes the peptide backbone improves resistance to exopeptidase attack, but the molecule stays susceptible to hydrolysis and oxidation once dissolved. Degradation accelerates with temperature, extreme pH, and light exposure, and repeated freeze-thaw cycles promote aggregation and material loss. Lyophilized powder held desiccated at or below minus twenty degrees Celsius is the common way to keep reference material. Reconstituted solutions are generally kept cold and used within a short window because their stability is far lower than that of the dry solid.
Handling procedures emphasize minimizing moisture uptake, since the dried solid is hygroscopic and can draw water when warmed to room temperature before a vial is opened. Laboratories record lot number, purity, counter-ion content, and residual solvent data supplied by the producer. Impurity profiles are compared run to run, and any shift in retention time or peak shape prompts re-verification against a reference standard. Certificates of analysis are treated as claims to be checked rather than accepted at face value.
Identity and purity testing for this peptide typically relies on reversed-phase high-performance liquid chromatography with ultraviolet detection, reported as area-percent purity. Mass spectrometry, usually in tandem mode, confirms molecular mass and supports quantification in biological matrices. Sequence confirmation may use peptide mapping after enzymatic digestion, while nuclear magnetic resonance and circular dichroism supply supplementary structural detail. No single technique establishes identity alone, so laboratories compare retention time, mass, and fragment pattern against a verified reference standard.
Across literature and commercial listings, the substance appears under several names, including bremelanotide, PT-141, and various brand designations. A CAS registry number provides a unique identifier for the compound, aiding retrieval from chemical databases. Synonym lists may also contain early development codes and descriptive chemical names. When consulting sources, researchers need to confirm that the cited names refer to the same molecular structure to avoid confusion. Naming consistency matters especially when comparing analytical data across publications.
PT-141 is a synthetic cyclic heptapeptide whose development code is bremelanotide. It belongs to the class of melanocortin receptor agonists and acts by mimicking endogenous peptide hormones. The compound originated from research on melanotan II, where investigators exploring derivatives found distinct pharmacological features. Unlike the parent compound, PT-141 showed effects on pathways related to sexual desire and function in early studies, prompting its development as a separate candidate. Its molecular design aimed to separate receptor activity from pigmentary effects.
Bremelanotide acts as a non-selective agonist at melanocortin receptors, with reported activity at MC1R, MC3R, MC4R and MC5R. The proposed basis for its central effects is activation of MC4R populations in the hypothalamus, a region associated with appetite and reproductive signalling. Because the peptide carries a net positive charge and polar side chains, it does not cross biological membranes freely, which is one reason oral administration is not the standard route. Effects generally appear within an hour of parenteral administration and are described as centrally mediated rather than peripheral.
Bremelanotide, developed under the code PT-141, is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone. Its structure is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, with a lactam bridge joining the aspartate and lysine side chains. The molecule has the formula C50H68N14O10 and a monoisotopic mass near 1025 daltons. It is commonly prepared as the acetate salt and appears as a white to off-white lyophilised powder in solid form. The free acid is the pharmacologically relevant species, while the counter-ion improves handling and dissolution.
In addition to cold seeps existing today, the fossil remains of ancient seep systems have been found in several parts of the world. Some of these are located far inland in places formerly covered by prehistoric oceans.
== Graphene analogs == Graphene analogs (also referred to as "artificial graphene") are two-dimensional systems which exhibit similar properties to graphene. Graphene analogs have been studied intensively since the discovery of graphene in 2004. People try to develop systems in which the physics is easier to observe and manipulate than in graphene. In those systems, electrons are not always the particles that are used. They might be optical photons, microwave photons, plasmons, microcavity polaritons, or even atoms. Also, the honeycomb structure in which those particles evolve can be of a different nature than carbon atoms in graphene. It can be, respectively, a photonic crystal, an array of metallic rods, metallic nanoparticles, a lattice of coupled microcavities, or an optical lattice.
low: hs-CRP level under 1.0 mg/L average: between 1.0 and 3.0 mg/L high: above 3.0 mg/L Normal levels increase with aging. Higher levels are found in late pregnant women, mild inflammation and viral infections (10–40 mg/L), active inflammation, bacterial infection (40–200 mg/L), severe bacterial infections and burns (>200 mg/L). CRP cut-off levels indicating bacterial from non-bacterial illness can vary due to co-morbidities such as malaria, HIV and malnutrition and the stage of disease presentation. In patients presenting to the emergency department with suspected sepsis, a CRP/albumin ratio of less than 32 has a negative predictive value of 89% for ruling out sepsis. CRP is a more sensitive and accurate reflection of the acute phase response than the ESR (erythrocyte sedimentation rate). ESR may be normal while CRP is elevated. CRP returns to normal more quickly than ESR in response to therapy.
== Structure and characteristics == Although many (>100) forms of pyoverdine have been isolated and studied, they all have certain characteristics in common. Each pyoverdine molecule has three parts: a dihydroxyquinoline core, a 6-14 amino acid peptide that varies among strains, and a side chain (usually composed of a 4-5 carbon α-ketoacid from the Krebs/citric acid cycle). The core of pyoverdine is responsible for several of its properties, including its well-known yellowish color and fluorescence.
=== Endothelial dysfunction and diabetic would healing === ITGA1 has been associated with vascular dysfunction in diabetic would healing, more specifically diabetic foot ulcers. Increased ITGA1 expression has been observed in diabetic wound tissues and AGEs-treated human umbilical vein endothelial cells (HUVECs). While elevated ITGA1 expression contributes to endothelial cell dysfunction by impairing proliferation, migration, and tube formation, while promoting cellular senescence; inhibiting ITGA1 restores endothelial cell function. This suggests ITGA1's contribution to diabetic wound pathology, and may represent a potential therapeutic target for vascular repair.

== Gene == The CTNS gene is located on the p arm of human chromosome 17, at position 13.2. It spans base pairs 3,636,468 and 3,661,542, and comprises 12 exons. In 1995, the gene was localized to the short arm of chromosome 17. An international collaborative effort finally succeeded in isolating CTNS by positional cloning in 1998. The CTNSN323K, CTNSK280R, and CTNSN288K mutations completely stop the movement of CySS out of the lysosome via cystinosin.[2] interestingly, CTNSN323K and CTNSK280R are related to juvenile nephropathic cystinosis while CTNSN288K mutations are found in cases with infantile nephropathic cystinosis.
and the Justice Department to investigate the contra-drug connection." Other journalists, including James Adams of The New York Times, were critical of Webb's work with his sources. Adams denounced Webb for his failure to contact the CIA and "cross check sources and allegations" and concluded, "For investigative reporters determined to uncover the truth, procedures like these are unacceptable." The series resulted in three federal investigations (by the CIA, the Department of Justice, and the House Intelligence Committee) into the claims of "Dark Alliance." The reports rejected his main claims but were critical of some CIA and law enforcement actions. The CIA report found no evidence that "any past or present employee of CIA, or anyone acting on behalf of CIA, had any direct or indirect dealing" with Ross, Blandón, or Meneses or that any of the other figures mentioned in "Dark Alliance" were ever employed by or associated with or contacted by the agency. The Department of Justice report stated that "We did not find that he [Blandón] had any ties to the CIA, that the CIA intervened in his case in any way, or that any connections to the Contras affected his treatment." The House Committee report examined the support that Meneses and Blandón gave to the local Contra organization in San Francisco and the Contras in general and concluded that it was "not sufficient to finance the organization" and did not consist of "millions," contrary to the claims of the "Dark Alliance" series.
== Legacy == In 1976, the National Park Service designated the Charles Richard Drew House in Arlington County, Virginia, as a National Historic Landmark in response to a nomination by the Afro-American Bicentennial Corporation. In 1981, the United States Postal Service issued a 35¢ postage stamp in its Great Americans series to honor Drew. In 2002, scholar Molefi Kete Asante listed Drew as one of the 100 Greatest African Americans.
=== Limitations to bioavailability === A recent study has shown Chlorambucil to be detoxified by human glutathione transferase Pi (GST P1-1), an enzyme that is often found over-expressed in cancer tissues. This is important since Chlorambucil, as an electrophile, is made less reactive by conjugation with glutathione, thereby making the drug less toxic to the cell.
=== Campaign finances reports === Many parties failed to report their campaign finances in a timely fashion, leading to suspicion over the use of the funds. On 21 November 2021, the ONPE sent auditors to supervise the management of party funds. The first parties scheduled to be audited are the Purple Party, Alliance for Progress, We Are Peru, and Popular Force. Later, Go on Country, Free Peru, and Together for Peru will be audited.

Poppy straw (also known as opium straw, mowed opium straw, crushed poppy capsule, poppy chaff, or poppy husk) is derived from opium poppies (Papaver somniferum) that are harvested when fully mature and dried by mechanical means. Opium poppy straw is what remains after the seed pods have been harvested — that is, the dried stalks, stem and leaves of poppies grown for their seeds. The field-dried leaves, stalk, and seed pod are then used in commercial manufacture of morphine or other poppy-alkaloid derived drugs, by first processing the material, separating the seeds, and then making concentrate of poppy straw where no extraction using the traditional methods of latex extraction has been made. The straw was originally considered an agricultural by-product of the mechanised poppy seed harvest, which was primarily grown for its edible and oil-producing seed. This changed in 1927 when János Kabay developed a chemical process to extract morphine from the crushed capsule. Concentrated poppy straw, consisting mainly of the crushed capsule without the seeds, soon became a valuable source of morphine. Today, concentrate of poppy straw is a major source of many opiates and other alkaloids. It is the source of 90% of the world supply of legal morphine (for medical and scientific use) and in some countries it also is a source of illegal morphine, which could be processed into illegal heroin. The 1961 Single Convention on Narcotic Drugs defines poppy straw as "all parts (except the seeds) of the opium poppy, after mowing".
== Side effects == Common side effects include nausea and vomiting, loss of appetite, change in taste, thin or brittle hair, pain in the joints of the arms or legs lasting two to three days, changes in the color of the nails, and tingling in the hands or toes. More serious side effects such as unusual bruising or bleeding, pain, redness or swelling at the injection site, hand-foot syndrome, change in normal bowel habits for more than two days, fever, chills, cough, sore throat, difficulty swallowing, dizziness, shortness of breath, severe exhaustion, skin rash, facial flushing, female infertility by ovarian damage, and chest pain can also occur. Neuropathy may also occur. Dexamethasone is given prior to paclitaxel infusion to mitigate some of the side effects. A number of these side effects are associated with the excipient used, Cremophor EL, a polyoxyethylated castor oil. Allergies to cyclosporine, teniposide, and other drugs delivered in polyoxyethylated castor oil may increase the risk of adverse reactions to paclitaxel.
=== Abnormality battles === Players can fight in Abnormality battles, boosting the respective floor's realization level and power. Many of the Abnormalities present in-game originate from the prequel, Lobotomy Corporation. Every Abnormality battle has a certain gimmick or mechanic the player must figure out to defeat them. After completing an Abnormality battle, a new librarian will be unlocked for the floor. Including the Patron Librarian, five librarians in total will be available per floor. Players can customize a librarian's name, appearance, and combat dialogue. Completing Abnormality battles also grants Abnormality pages used in battles. The Abnormality pages can be split into two categories: Awakening and Breakdown. The Awakening Abnormality cards are the green ones and provide a small benefit with no downsides. The Breakdown Abnormality cards are red, with both a strong benefit and a downside. The severity of the downside depends on the card itself.

PT-141 is a laboratory code for bremelanotide, a synthetic peptide that activates melanocortin receptors. It has been developed both as an injectable pharmaceutical product and as a research chemical. The two contexts differ sharply in how identity, purity, and dosing are controlled.
It is neither a steroid nor a naturally occurring human hormone. Bremelanotide is a manufactured peptide that mimics part of the melanocortin signalling system by binding receptor proteins. Its effects come from receptor activation, not from replacing a circulating hormone.
The internal lactam bridge locks the peptide into a defined shape, which raises receptor affinity and slows enzymatic breakdown. Linear versions of the same sequence are less stable and bind more weakly. Ring geometry is therefore central to how the molecule behaves in the body.
Trial results generally show a small to moderate average improvement in desire scores relative to placebo. The distribution of responses is wide, and some participants show little measurable change. Group averages should not be read as a prediction for any single person.
