=== Professional ski wax technicians === Compared to the general public exposed to contaminated drinking water, professional ski wax technicians are more strongly exposed to PFAS (PFOA, PFNA, PFDA, PFHpA, PFDoDA) from the glide wax used to coat the bottom of skis to reduce the friction between the skis and snow. During the coating process, the wax is heated, which releases fumes and airborne particles. Compared to all other reported occupational and residential exposures, ski waxing had the highest total PFAS air concentrations.
Some longitudinal incisions of 2mm of depth have to be made through the skin to proceed to the extraction of latex, which can be collected in solid form some minutes after the extraction. The proteases present in the latex of the fruit are inactive precursors that are activated once the papaya is wounded. In 0.3 ml of latex there are about 15 mg of chymopapain. If we want to conserve the proteolytic properties, latex has to be preserved with sodium metabisulfite and stored at a low temperature of about -10 °C. If used immediately after the incisions, a buffer is added to extract the proteins: EDTA, ammonium sulfate or phosphate buffer all with a concentration of 0.5 mM and a pH of 7. It is also important to block the thiol functions to avoid air oxidation and the loss of proteolytic activity. To eliminate organic and insoluble molecules, the sample is first filtered and afterwards centrifuged at 11000g for 30min. The pellet is discarded and the supernatant added to 96% alcohol with a ratio of 1:3. Impurities precipitate and can be eliminated by filtration. Afterwards, (NH4)2SO4 fractioning is done by addition of this substance at a concentration of 0.472 mg/ml. Chymopapain precipitates and can be retrieved through another centrifugation, again at 11000g for 30min. The supernatant is discarded and the ion exchange chromatography can be carried out, with a linear gradient of 100mM (Na+) and different volumes of elution. === Candidacy for mayor of Porto Alegre in 2020 === In June 2020, Melo launched his pre-candidacy to run for mayor again, bolstered by a unanimous intra-party vote. His vice-mayoral candidate was Ricardo Gomes (DEM). Other members of his coalition included Cidadania, Solidariedade, Christian Democracy, and the PRTB. Despite the PSL, the former party of former president Jair Bolsonaro, officially supporting Marchezan Júnior's reelection campaign, some deputies from the party campaigned for Melo. In the first round, he received 200,280 votes (31%), and advanced to the second round against Manuela d'Ávila, who received 187,262 (29%). Extending the time during which the cider remains in contact with yeast lees increased concentrations of most of the minor volatile compounds present, especially fatty acids, ethyl esters and alcohols. Major volatile compound concentrations did not exhibit a similar pattern, with iso-butanol, amyl alcohols, and acetone decreasing 1-propanol decreasing. Sparkling ciders can be produced using different methods, including the Champenoise method used to produce champagne. Use of different strains of indigenous Saccharomyces to perform secondary fermentation produced ciders with consistent alcohol and acidic characteristics, variable glycerol, acetaldehyde, ethyl acetate, methanol, propanol, i-butanol and 2-phenylethanol characteristics and acceptable sensory analysis results.
