Development of the peptide passed through several delivery formats, including an intranasal version tested in early trials and an injectable version that entered later clinical study. Regulatory approval for a subcutaneous product in the United States was granted in 2019 after review of controlled trials in premenopausal women. Outside the clinic, the compound circulates in research and non-pharmaceutical markets under its code name, where identity and purity vary considerably between suppliers. Synonyms appearing across technical literature include bremelanotide, PT-141, and Palatin 141.
PT-141 is the research designation for bremelanotide, a synthetic cyclic heptapeptide developed as a melanocortin receptor agonist. The peptide contains seven amino acids with an internal lactam bridge that constrains the backbone into a stable ring. Cyclisation of this kind improves resistance to enzymatic degradation relative to linear analogues. The compound was first explored for effects on melanin production, because several melanocortin receptors influence pigmentation. Its later association with sexual desire emerged from observations made during that early work, and the molecule became the subject of a separate development programme.
Clinical research typically uses randomised, double-blind, placebo-controlled designs. The most common primary endpoint is the desire domain score of the Female Sexual Function Index, sometimes paired with a distress measure. Secondary outcomes include arousal, satisfaction, and event-based counts of satisfying sexual episodes. Across trials, average improvements are modest and individual responses vary widely. Whether benefits persist beyond a few months, and whether they depend on baseline hormone status, remain open questions rather than settled findings.
Melanocortin receptors form a family of five G-protein-coupled receptors designated MC1 through MC5. Bremelanotide binds most strongly at MC4R and MC1R, with weaker activity reported at MC3R and MC5R. MC4R is expressed in hypothalamic nuclei that coordinate energy balance and aspects of sexual behaviour. The prevailing interpretation is that central MC4R activation, rather than peripheral vascular effects alone, drives the reported changes in desire. This account remains partly inferential, since direct receptor-level measurement in living humans is not practical.

Receptor activation in hypothalamic and limbic circuits is the mechanism most often cited for the observed effects on sexual desire. Signalling through MC4R couples to Gs proteins and raises intracellular cyclic AMP, which in turn modulates dopaminergic tone in reward-related pathways. Because the peptide reaches the central nervous system after subcutaneous administration, peripheral vascular changes are regarded as secondary rather than primary. The precise neural circuits that translate receptor occupancy into behavioural change remain incompletely mapped, and published accounts describe the pathway in general terms rather than as a fully resolved sequence.
Development began with intranasal formulations investigated for erectile dysfunction, but blood pressure elevation limited that route and prompted a switch to subcutaneous delivery. Clinical testing then shifted toward hypoactive sexual desire disorder in premenopausal women, and a subcutaneous product received United States approval in 2019. Later trials examined other populations with mixed results, and questions about effect size, durability and patient selection remain open in the peer-reviewed literature. Research interest continues in parallel with the broader melanocortin field, where several synthetic analogues are studied together.
PT-141 is the research code for bremelanotide, a cyclic heptapeptide derived from alpha-melanocyte-stimulating hormone. The molecule belongs to the melanocortin receptor agonist family and shows highest affinity for the MC4 receptor subtype, with weaker activity at MC1, MC3 and MC5. Its structure retains the core His-Phe-Arg-Trp sequence that defines melanocortin recognition, while cyclization and terminal modifications improve metabolic stability relative to the parent hormone. Early work classified the compound as a centrally acting agent rather than a peripherally acting vasodilator, which shaped subsequent development priorities.
Handling procedures emphasize minimizing moisture uptake, since the dried solid is hygroscopic and can draw water when warmed to room temperature before a vial is opened. Laboratories record lot number, purity, counter-ion content, and residual solvent data supplied by the producer. Impurity profiles are compared run to run, and any shift in retention time or peak shape prompts re-verification against a reference standard. Certificates of analysis are treated as claims to be checked rather than accepted at face value.
Identity and purity testing for this peptide typically relies on reversed-phase high-performance liquid chromatography with ultraviolet detection, reported as area-percent purity. Mass spectrometry, usually in tandem mode, confirms molecular mass and supports quantification in biological matrices. Sequence confirmation may use peptide mapping after enzymatic digestion, while nuclear magnetic resonance and circular dichroism supply supplementary structural detail. No single technique establishes identity alone, so laboratories compare retention time, mass, and fragment pattern against a verified reference standard.
Across literature and commercial listings, the substance appears under several names, including bremelanotide, PT-141, and various brand designations. A CAS registry number provides a unique identifier for the compound, aiding retrieval from chemical databases. Synonym lists may also contain early development codes and descriptive chemical names. When consulting sources, researchers need to confirm that the cited names refer to the same molecular structure to avoid confusion. Naming consistency matters especially when comparing analytical data across publications.
PT-141 is a synthetic cyclic heptapeptide whose development code is bremelanotide. It belongs to the class of melanocortin receptor agonists and acts by mimicking endogenous peptide hormones. The compound originated from research on melanotan II, where investigators exploring derivatives found distinct pharmacological features. Unlike the parent compound, PT-141 showed effects on pathways related to sexual desire and function in early studies, prompting its development as a separate candidate. Its molecular design aimed to separate receptor activity from pigmentary effects.
Bremelanotide acts as a non-selective agonist at melanocortin receptors, with reported activity at MC1R, MC3R, MC4R and MC5R. The proposed basis for its central effects is activation of MC4R populations in the hypothalamus, a region associated with appetite and reproductive signalling. Because the peptide carries a net positive charge and polar side chains, it does not cross biological membranes freely, which is one reason oral administration is not the standard route. Effects generally appear within an hour of parenteral administration and are described as centrally mediated rather than peripheral.
Bremelanotide, developed under the code PT-141, is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone. Its structure is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, with a lactam bridge joining the aspartate and lysine side chains. The molecule has the formula C50H68N14O10 and a monoisotopic mass near 1025 daltons. It is commonly prepared as the acetate salt and appears as a white to off-white lyophilised powder in solid form. The free acid is the pharmacologically relevant species, while the counter-ion improves handling and dissolution.
Early research on PT-141 grew out of work on melanotan II, a related cyclic peptide studied for pigmentation. Investigators observed that centrally acting melanocortin agonists also influenced sexual behaviour in animal models, and the programme shifted toward that endpoint. A nasal formulation was evaluated in clinical trials but showed inconsistent absorption, and later studies used subcutaneous administration instead. Regulatory approval in the United States followed in 2019 for a defined population of premenopausal women with acquired, generalised hypoactive sexual desire disorder. That approval was specific to that group rather than a broad indication.
. This shows that the density of the material is an important factor when designing structures that can withstand loads, and that the Young's modulus of the structure is heavily determined by the porosity of the structure. Past the linear region, the lamellae start to buckle elastically and deform non-linearly. In a stress-strain curve, this is shown as a flat plateau. The critical load at which buckling begins is given by:
== Natural history == Although it is generally accepted that vanilla was domesticated in Mesoamerica and subsequently spread to the Old World in the 16th century, in 2019, researchers published a paper stating that vanillin residue had been discovered inside jars within a tomb in Canaan dating to the 2nd millennium BCE, suggesting the possible cultivation of an unidentified, Old World-endemic Vanilla species in Canaan since the Middle Bronze Age. Traces of vanillin were also found in wine jars in Jerusalem, which were used by the Judahite elite before the city was destroyed in 586 BCE. Vanilla beans, called tlilxochitl, were discovered and cultivated as a flavoring for beverages by native Mesoamerican peoples, most famously the Totonacs of modern-day Veracruz, Mexico. Since at least the early 15th century, the Aztecs used vanilla as a flavoring for chocolate in drinks called xocohotl.
=== Burial at crossroads === Historically, crossroad graves were used to dispose of the bodies of executed criminals and suicides. They were located at a crossroads, often on a parish border. In Great Britain, until the Burial of Suicides Act 1823, suicides were generally not allowed a burial in consecrated ground, and the burial far outside the community, sometimes with a stake through their heart, was seen as a way to keep their spirits from haunting the area. Crossroads form a crude cross shape and this may have given rise to the belief that these spots were selected as burying-places which were next best to consecrated ground. The shape was seen as a powerful symbol against all kinds of unwelcome powers. Another possible explanation is that the ancient Teutonic (Germanic) ethnic groups often built their altars at a crossroads, and since human sacrifices, especially of criminals, formed part of ritual, these spots came to be regarded as execution grounds. Hence after the introduction of Christianity, criminals and suicides were buried at the crossroads during the night, to assimilate as far as possible their funeral with that of the pagans. An example of a crossroad execution-ground was the famous Tyburn in London, which stood on the spot where the Roman road to Edgware and beyond met the Roman road heading west out of London. Superstition also played a part in the selection of crossroads in the burial of suicides.
== Epidemiology and prevention == Selenium deficiency is uncommon, but regions in China, Europe, Russia, and New Zealand have low selenium levels in croplands and diet. The worldwide prevalence of selenium deficiency is however predicted to rise under climate change due to the loss of selenium from croplands. These diseases are most common in certain parts of China where the intake is low because the soil is extremely deficient in selenium. Studies in Jiangsu Province of China have indicated a reduction in the prevalence of these diseases by taking selenium supplements. In Finland, selenium salts are added to chemical fertilizers, as a way to increase selenium in soils. Dietary supplements may utilize sodium selenite, L-selenomethionine, or selenium-enriched yeast.
== Prime Minister of Thailand (2024–2025) == Following the removal of Srettha as prime minister by the Constitutional Court of Thailand on 14 August 2024, Paetongtarn was nominated by Pheu Thai to succeed him. Her nomination was approved by the House of Representatives on 16 August after no alternatives were named by the other parties in the ruling coalition, making her the youngest person and the second woman to become Prime Minister of Thailand. She was officially sworn in on 18 August following an endorsement from King Vajiralongkorn, followed by her cabinet on 6 September. Her premiership has been plagued by the return of her father Thaksin Shinawatra, a prominent and highly controversial political figure of Thailand. This led to a vote of no-confidence by the opposition, alleging that she has allowed her father to control the Thai government and does not have her own autonomy to run the country – Paetongtarn survived this and remained in position.

Edible insects are raised as livestock in specialized insect farms. In North American as well as European countries such as the Netherlands or Belgium, insects are produced under strict food law and hygiene standards for human consumption. Conditions such as temperature, humidity, feed, water sources, and housing, vary depending on the insect species. The insects are raised from eggs to larvae status (mealworms, lesser mealworms) or to their mature form (crickets, locusts) in industrialized insect farms and then killed via temperature control. Culled insects may be freeze-dried and packed whole, or pulverized to insect powder (insect flour) to be used in other food products such as baked goods or snacks. In addition to nutritional composition and digestibility, insect species are selected for ease of rearing by the producer based on factors such as disease susceptibility, feed conversion efficiency, rate of development, and generational turnover.
autophagy Also autophagocytosis. The orderly autolytic degradation and recycling of dysfunctional or unnecessary cellular components by the cell's own enzymes as part of a carefully regulated, lysosome-dependent pathway. Autophagic programs play important roles in nutrient-deprived and senescent cells but also help maintain homeostasis in healthy cells.
In February 1917, revolution broke out in Russia in which workers, soldiers and peasants established soviets, the monarchy was forced into exile fell and a provisional government was formed until the election of a constituent assembly. Alexander Kerensky, a Russian lawyer and revolutionary, became a key political figure in the Russian Revolution of 1917. After the February Revolution, Kerensky joined the newly formed Russian Provisional Government, first as Minister of Justice, then as Minister of War and after July as the government's second Minister-Chairman. A leader of the moderate socialist Trudovik faction of the Socialist Revolutionary Party known as the Labour Group, Kerensky was also the vice-chairman of the powerful Petrograd Soviet. After failing to sign a peace treaty with the German Empire to exit from World War I which led to massive popular unrest against the government cabinet, Kerensky's government was overthrown on 7 November by the Bolsheviks led by Vladimir Lenin in the October Revolution. Soon after the October Revolution, the Russian Constituent Assembly elected Socialist-Revolutionary leader Victor Chernov as President of a Russian Republic, but it rejected the Bolshevik proposal that endorsed the Soviet decrees on land, peace and workers' control and acknowledged the power of the Soviets of Workers', Soldiers' and Peasants' Deputies.
Training in internal medicine (as opposed to surgical training), varies considerably across the world: see the articles on medical education for more details. In North America, it requires at least three years of residency training after medical school, which can then be followed by a one- to three-year fellowship in the subspecialties listed above. In general, resident work hours in medicine are less than those in surgery, averaging about 60 hours per week in the US. This difference does not apply in the UK where all doctors are now required by law to work less than 48 hours per week on average.

=== Excessive sleepiness === Selegiline has been evaluated for the treatment of narcolepsy in three small clinical studies. It was found to be effective in these studies. A dosage of 10 mg/day had no effect on symptoms, but 20 to 30 mg/day improved alertness, mood, and somewhat reduced cataplexy, clinical effects that have been described as comparable to the same dosages of amphetamine. Animal research indicates that the beneficial effects of high doses of selegiline in narcolepsy are likely due to conversion into its active metabolites, levoamphetamine and levomethamphetamine. Selegiline has also been evaluated for treatment of hypersomnia (excessive sleeping or sleepiness) in people with myotonic dystrophy, but its effectiveness was very uncertain.
=== Lebanon === In June 2025 Lebanese authorities uncovered a major criminal network that smuggled and sold counterfeit cancer drugs, including fake immunotherapy medication like IMFINZI (durvalumab), through Beirut's airport. These medicines contained no active ingredients and were substituted for real chemotherapy drugs, placing patients' lives at risk. The judicial inquiry, led by Judge Dora al-Khazen, has resulted in at least five arrests, including Mohamad Khalil (brother of former Finance Minister Ali Hassan Khalil), his ex-wife (a Beirut-area pharmacist), and a security officer who facilitated the smuggling. The scandal triggered emergency governmental action, widespread pharmacy raids, and significant public outrage, as lawmakers warned that up to one-third of medicines in Lebanon could be fake.
Studies in biochemistry show that endogenous, bacterial chaperones of curli are capable of briefly interacting with Syn and controlling its aggregation. The clinical and pathological findings support the hypothesis that aSyn disease in PD occurs via a gut-brain pathway. For early diagnosis and early management in the phase of creation and propagation of aSyn, it is therefore of utmost importance to identify pathogenic aSyn in the digestive system, for example, by gastrointestinal tract (GIT) biopsies. According to a growing body of research, intestinal dysbiosis may be a major factor in the development of Parkinson's disease by encouraging intestinal permeability, gastrointestinal inflammation, and the aggregation and spread of asyn. Not just the CNS but other peripheral tissues, such as the GIT, have physiological aSyn expression as well as its phosphorylated variants. As suggested by Borghammer and Van Den Berge (2019), one approach is to recognise the possibility of PD subtypes with various aSyn propagation methods, including either a peripheral nervous system (PNS)-first or a CNS-first route. While the GI tract has been linked to other neurological disorders such autism spectrum disorder, depression, anxiety, and Alzheimer's disease, protein aggregation and/or inflammation in the gut represent a new topic of investigation in synucleinopathies.
Yet, because the Cortes of Cádiz was located in Spain, political and economic power and decisions were localized in Spain, effectively giving them control over all of colonial Spanish America. These tensions further frustrated many Spanish-Americans because of their inability to control the politics that directly affected their economic and sociopolitical wellbeing, further leading them towards independence. This climate of instability created the conditions for the two sides to forge an alliance. This alliance coalesced towards the end of 1820 behind Agustín de Iturbide, a colonel in the royal army, who at the time was assigned to destroy the guerrilla forces led by Vicente Guerrero.
Both sodium and potassium are commonly used as GRAS counterions to create more water-soluble and hence more bioavailable salt forms of acidic pharmaceuticals. Potassium compounds are often used as fertilisers as potassium is an important element for plant nutrition. Potassium hydroxide is a very strong base, and is used to control the pH of various substances. Potassium nitrate and potassium permanganate are often used as powerful oxidising agents. Potassium superoxide is used in breathing masks, as it reacts with carbon dioxide to give potassium carbonate and oxygen gas. Pure potassium metal is not often used, but its alloys with sodium may substitute for pure sodium in fast breeder nuclear reactors. Rubidium and caesium are often used in atomic clocks. Caesium atomic clocks are extraordinarily accurate; if a clock had been made at the time of the dinosaurs, it would be off by less than four seconds (after 80 million years). For that reason, caesium atoms are used as the definition of the second. Rubidium ions are often used in purple fireworks, and caesium is often used in drilling fluids in the petroleum industry. Francium has no commercial applications, but because of francium's relatively simple atomic structure, among other things, it has been used in spectroscopy experiments, leading to more information regarding energy levels and the coupling constants of the weak interaction. Studies on the light emitted by laser-trapped francium-210 ions have provided accurate data on transitions between atomic energy levels, similar to those predicted by quantum theory.

PT-141 is a laboratory code for bremelanotide, a synthetic peptide that activates melanocortin receptors. It has been developed both as an injectable pharmaceutical product and as a research chemical. The two contexts differ sharply in how identity, purity, and dosing are controlled.
It is neither a steroid nor a naturally occurring human hormone. Bremelanotide is a manufactured peptide that mimics part of the melanocortin signalling system by binding receptor proteins. Its effects come from receptor activation, not from replacing a circulating hormone.
The internal lactam bridge locks the peptide into a defined shape, which raises receptor affinity and slows enzymatic breakdown. Linear versions of the same sequence are less stable and bind more weakly. Ring geometry is therefore central to how the molecule behaves in the body.
Trial results generally show a small to moderate average improvement in desire scores relative to placebo. The distribution of responses is wide, and some participants show little measurable change. Group averages should not be read as a prediction for any single person.
