The melanocortin system comprises five G protein-coupled receptors, designated MC1 through MC5, that signal mainly through cyclic AMP accumulation. MC1R and MC2R are associated with pigmentation and adrenal steroid production, while MC3R and MC4R are expressed in the central nervous system and influence energy balance and behavior. MC5R appears in exocrine tissues. Natural agonists include alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone, and endogenous antagonists such as agouti-related protein modulate the same sites. This receptor family provides the framework within which bremelanotide activity is described.
Bremelanotide acts as an agonist at several melanocortin receptors, with the strongest reported activity at MC4R and measurable activity at MC1R and MC3R. Because MC4R is expressed in hypothalamic and limbic circuits, the proposed mechanism links receptor activation to modulation of central pathways involved in desire rather than to direct effects on peripheral genital tissue. The precise downstream steps remain incompletely characterized, and evidence for the involvement of specific neurotransmitters is suggestive rather than settled. Nausea and blood pressure elevation reported during trials are consistent with melanocortin signaling outside the intended target circuit.
Compared with melanotan II, bremelanotide is a smaller cyclic peptide with a more constrained backbone, which affects receptor selectivity and metabolic stability. Published descriptions give a plasma half-life on the order of a few hours after subcutaneous administration, with elimination through hepatic and renal routes and limited plasma protein binding. Central access is inferred from effects observed in animal models, although direct measurement in humans is limited. Handling and storage requirements follow from the peptide backbone, which is susceptible to hydrolysis and oxidation.
Early research on PT-141 grew out of work on melanotan II, a related cyclic peptide studied for pigmentation. Investigators observed that centrally acting melanocortin agonists also influenced sexual behaviour in animal models, and the programme shifted toward that endpoint. A nasal formulation was evaluated in clinical trials but showed inconsistent absorption, and later studies used subcutaneous administration instead. Regulatory approval in the United States followed in 2019 for a defined population of premenopausal women with acquired, generalised hypoactive sexual desire disorder. That approval was specific to that group rather than a broad indication.
Bremelanotide acts as a non-selective agonist at melanocortin receptors, with reported activity at MC1R, MC3R, MC4R and MC5R. The proposed basis for its central effects is activation of MC4R populations in the hypothalamus, a region associated with appetite and reproductive signalling. Because the peptide carries a net positive charge and polar side chains, it does not cross biological membranes freely, which is one reason oral administration is not the standard route. Effects generally appear within an hour of parenteral administration and are described as centrally mediated rather than peripheral.
Bremelanotide, developed under the code PT-141, is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone. Its structure is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, with a lactam bridge joining the aspartate and lysine side chains. The molecule has the formula C50H68N14O10 and a monoisotopic mass near 1025 daltons. It is commonly prepared as the acetate salt and appears as a white to off-white lyophilised powder in solid form. The free acid is the pharmacologically relevant species, while the counter-ion improves handling and dissolution.

Bremelanotide is a synthetic cyclic heptapeptide that acts on a family of G-protein-coupled receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide associated with pigmentation and several central signalling pathways. A lactam bridge constrains the ring and slows enzymatic breakdown, which distinguishes it from the linear parent molecule. Research interest moved over time from pigment biology toward central nervous system effects, particularly circuits connected to sexual desire. Parenteral delivery is used because oral bioavailability is poor.
Early clinical work used an intranasal formulation, and later programmes switched to subcutaneous delivery for more consistent absorption. A subcutaneous product received regulatory approval in the United States in 2019 for premenopausal women with acquired, generalised hypoactive sexual desire disorder. Approval followed phase 3 trials in which active treatment separated from placebo on desire and distress measures, though the average difference was modest. Labeling carries a caution about transient blood pressure elevation, so cardiovascular history is assessed before prescribing. Questions about durability of benefit beyond several months remain open.
Published discussion sits at the intersection of peptide chemistry, neuroendocrinology and sexual medicine. Trial reports emphasise change scores on validated instruments, while mechanistic papers focus on hypothalamic circuits and receptor selectivity. Because placebo response in this field is large, effect sizes are usually reported with confidence intervals rather than as isolated averages. Reviews note that female and male data sets are not interchangeable and should be read separately. Diagnostic terminology has been revised over time, which complicates comparison between older and newer studies.
Gas exchange must be restored as quickly as possible to avoid collateral damage, so activated lymphocytes secrete IFNγ to stimulate the production of matrix metalloproteinase MMP-9 by macrophages. AMs have been reported to produce MMP-9 partly via PGE2-dependent PKA signaling pathways, which are the pathways involved in the inhibition of phagocytosis. MMP-9 activates latent TGF-β, reinducing expression of αvβ6 integrins on alveolar epithelial cells, thereby returning the alveolar macrophage to a resting state. Activation of TGF-β is also advantageous because its production stimulates collagen synthesis in interstitial fibroblasts, which is necessary for restoring alveolar wall architecture. In a November 2004 interview, Boyarsky said that although the team would like to pursue a Bloodlines sequel, the decision was Activision's. Before their closure, Troika had begun development of a workable prototype based on another of White Wolf's tabletop role-playing games, Werewolf: The Apocalypse, set in the same universe as Vampire: The Masquerade. According to Boyarsky, the prototype was one small area built using assets taken from Bloodlines, and allowed the player to play as a werewolf, or a human capable of turning into one. Paradox Interactive obtained the rights to Bloodlines in 2015, following their purchase of White Wolf. Paradox CEO Fredrik Wester confirmed that a sequel was possible, stating "when the time is right I guess a sequel will find its place in the market." Vampire: The Masquerade – Bloodlines 2 was announced in March 2019. Initially developed by Seattle-based Hardsuit Labs, the game was to be written by Bloodlines's lead-writer Brian Mitsoda, and Cara Ellison. Set in Seattle, the plot cast the player as a fledgling thinblood vampire with relatively weak abilities, who was turned as part of a masquerade-violating "mass embrace" of humans. However, in 2020, Mitsoda, Ellison, and creative director Ka'ai Cluney left the project. In February 2021, Hardsuit Labs was also removed as developer and replaced with The Chinese Room. The change also resulted in a different plot which follows an elder vampire, Phyre, after they are awoken in modern Seattle. The sequel was released on 21 October 2025. == Structure-activity relationship == The structure-activity relationship (SAR) of gliflozins is not fully understood. The most commonly used gliflozins are dapagliflozin, empagliflozin, and canagliflozin. The differences in the structures are relatively small. The general structure includes a glucose sugar with an aromatic group in the β-position at the anomeric carbon. In addition to the glucose sugar moiety and the β-isomeric aryl substituent, the aryl group is composed of a diarylmethylene structure. The synthesis of gliflozins involves three general steps. The first one is the construction of the aryl substituent, the next one is the introduction of the aryl moiety onto the sugar or glucosylation of the aryl substituent, and the last one is the deprotection and modification of the arylated anomeric center of the sugar. Phlorizin was the first type of gliflozin, and it was non-selective against SGLT2/SGLT1. It is a natural O-aryl glycoside composed of a d-glucose and an aromatic ketone. However phlorizin is very unstable, it is rapidly degraded by glucosidases in the small intestines, so it can not be used as an orally administrered medication to treat diabetes. Structural modifications have been made to overcome this instability problem. The most efficient way was to conjugate the aryl moiety with the glucose moiety since C-glucosides are more stable in the small intestines than O-glucoside derivatives (C-C bond instead of C-O-C bond).
Glucose + 2 NAD+ + 2 Pi + 2 ADP → 2 pyruvate + 2 NADH + 2 ATP + 2 H+ + 2 H2O + energy Starting with glucose, 1 ATP is used to donate a phosphate to glucose to produce glucose 6-phosphate. Glycogen can be converted into glucose 6-phosphate as well with the help of glycogen phosphorylase. During energy metabolism, glucose 6-phosphate becomes fructose 6-phosphate. An additional ATP is used to phosphorylate fructose 6-phosphate into fructose 1,6-bisphosphate by the help of phosphofructokinase. Fructose 1,6-biphosphate then splits into two phosphorylated molecules with three carbon chains which later degrades into pyruvate.

== Purification of plasmid DNA == It is important to consider the downstream applications of the plasmid DNA when choosing a purification method. For example, if the plasmid is to be used for transfection or electroporation, a purification method that results in high purity and low endotoxin levels is desirable. Similarly, if the plasmid is to be used for sequencing or PCR, a purification method that results in high yield and minimal contaminants is desirable. However, multiple methods of nucleic acid purification exist. All work on the principle of generating conditions where either only the nucleic acid precipitates, or only other biomolecules precipitate, allowing the nucleic acid to be separated.
A U.S. company named VAZTEC has created a compliant rotary valve sealing system that solves the friction and sealing problems of previous designs. They have built ten prototype engines from a 5.3L V-8 to a handheld 28cc four stroke. VAZTEC has also built a successful Diesel engine that contained the high compression pressures of a compression ignition engine (20:1 compression ratio, 100 bar combustion pressure). They are working with various OE manufacturers to commercialize their design. Various patents cover the compliant seal, including U.S. Patent 9,903,239. A Computational Fluid Dynamics (CFD) model of the VAZTEC rotary valve can be viewed at this link.
=== Colorado === On May 17, 2024, Colorado Governor Jared Polis signed the Colorado AI Act (CAIA) into law. The CAIA establishes requirements for deployers and developers of high-risk AI systems, including transparency obligations and risk assessments, to prevent algorithmic discrimination against Colorado residents. It is the first comprehensive, state-level law that regulates high-risk AI systems in the United States. The law was unpopular among pro-business advocates. An attempt to amend the regulation failed, but its implementation was delayed from the original February 1, 2026 start date to an expected June 1, 2026 start. Jared Polis has created a committee to investigate its implementation. Brianna Titone, the bill's sponsor, criticized the commission for being full of industry representatives without representation from consumers across the state. On May 14, 2026, Polis signed into law SB 26-189, which repealed and replaced CAIA. The replacement law eliminated the focus on high-risk systems; instead, it requires companies that deploy AI for decision-making related to certain topics (like employment or housing) to inform individuals that AI is being used.

The sclera forms the posterior five-sixths of the connective tissue coat of the human eyeball. It is continuous with the dura mater and the cornea, and maintains the shape of the eyeball, offering resistance to internal and external forces, and provides an attachment for the extraocular muscle insertions. The sclera is perforated by many nerves and vessels passing through the posterior scleral foramen, the hole that is formed by the optic nerve. At the optic disc, the outer two-thirds of the sclera continues with the dura mater (outer coat of the brain) via the dural sheath of the optic nerve. The inner third joins with some choroidal tissue to form a plate (lamina cribrosa) across the optic nerve with perforations through which the optic fibers (fasciculi) pass. The thickness of the sclera varies from 1 mm at the posterior pole to 0.3 mm just behind the insertions of the four rectus muscles. The sclera's blood vessels are mainly on the surface. Along with the vessels of the conjunctiva (which is a thin layer covering the sclera), those in the episclera render the inflamed eye bright red. In many vertebrates, the sclera is reinforced with plates of cartilage or bone, together forming a circular structure called the scleral ring. In primitive fish, this ring consists of four plates, but the number is lower in many living ray-finned fishes, and much higher in lobe-finned fishes, crocodilians, various reptiles, and birds. The ring has disappeared in many groups, including living amphibians, some reptiles and fish, and all mammals.
β-Hydroxy β-methylbutyric acid is a monocarboxylic β-hydroxy acid and natural product with the molecular formula C5H10O3. At room temperature, pure β-hydroxy β-methylbutyric acid occurs as a transparent, colorless to light yellow liquid which is soluble in water. β-Hydroxy β-methylbutyric acid is a weak acid with a pKa of 4.4. === Reggio revolt === The Reggio revolt came on 14 July 1970, supported across social classes (bourgeoisie, clergy, students, political parties, civic committees). Clashes with authorities left one dead (railroad worker Bruno Labate), prompting Archbishop Vincenzo Ferro to join the showdown. The uprising was supported by liberal-conservative newspapers (such as Gazzetta del Sud and Il Tempo). Gradually protest leadership passed from Mayor Battaglia, to more assertive movements, particularly the Movimento Sociale Italiano, seen as least compromised with the republican regime. The Missini authored slogans (famous was the boia chi molla of D'Annunzian memory). Ciccio Franco, a CISNAL trade unionist and Reggio Calabria-based Missini exponent, emerged as the undisputed leader. Barricades were erected, the railway station was occupied and convoys and ferries to Sicily were blocked. Nineteen days of general strikes, 12 bomb attacks, 32 roadblocks, 14 occupations of the station, 2 of the post office, 1 of the television station, and 4 assaults on the prefecture, The death toll reached 5 on both sides (in addition to Labate, Angelo Campanella, also perished in the clashes, Vincenzo Curigliano, Antonio Bellotti, and Carmelo Jaconis). 426 were arrested and 200 were wounded during police charges (whose members were insulted and vilified even by hospital doctors). In some parts of the city, “autonomous republics” were proclaimed, such as the “Republic of Sbarre” and the “Grand Duchy of St. Catherine”.

Users can also use portions of other TikTok videos by creating a "stitch". TikTok announced a "family safety mode" in February 2020 for parents to be able to control their children's presence on the app. There is a screen time management option, restricted mode, and the option to put a limit on direct messages. The app expanded its parental controls feature called "Family Pairing" in September 2020 to provide parents and guardians with educational resources to understand what children on TikTok are exposed to. Content for the feature was created in partnership with online safety nonprofit, Internet Matters. In October 2021, TikTok launched a test feature that allows users to directly tip certain creators. Accounts of users that are of age, have at least 100,000 followers and agree to the terms can activate a "Tip" button on their profile, which allows followers to tip any amount, starting from $1. In December 2021, TikTok started beta-testing Live Studio, a streaming software that would let users broadcast applications open on their computers, including games. The software also launched with support for mobile and PC streaming. However, a few days later, users on Twitter discovered that the software uses code from the open-source OBS Studio. OBS made a statement saying that, under the GNU GPL version 2, TikTok has to make the code of Live Studio publicly available if it wants to use any code from OBS.

Reported activity is highest at MC4R, with lower potency at MC1R and MC3R. The MC4R interaction is generally treated as the most relevant to its central effects. Selectivity is not absolute, and activity across the family is dose-dependent.
Indirect evidence from animal studies supports central access, and the proposed mechanism requires it. Direct quantification in humans is limited. How much reaches specific brain regions remains an open question.
Published descriptions give a plasma half-life of roughly two to three hours after subcutaneous administration. Values vary with assay method and study population. The figure is an average rather than a fixed molecular property.
PT-141 was the development code used for bremelanotide during its preclinical and early clinical programme. The peptide is now generally referred to by its international nonproprietary name.
